Description of Fibronectin Adhesion-promoting Peptide

Fibronectin Adhesion-promoting Peptide(Trp-Glu-Pro-Pro-Arg-Ala-Arg-Ile) ist medizinisch interessant und hat auch eine gut charakterisierte Struktur. Aus medizinischer Sicht wurde die Sequenz(WQPPRARI) in der carboxyterminierten Heparin-Bindungsdomäne der Fibronectin-Repeats III14 gefunden. Es ist ein potenter Induktor von Stressfasern und fokaler Adhäsion in Fibroblasten, die an der Thrombosebildung im Zusammenhang mit Erkrankungen wie atherosclerotischen Herz-Kreislauf-Erkrankungen beteiligt sind.[2]

Fibronectin Adhesion-promoting Peptide hat ein niedriges Molekulargewicht von 1114.25 g/mol und hat struktuell keine Sekundärstuktur. Deshalb ist eine irreversible Adsorption aufgrund oberflächeninduzierter Konformationsänderungen voraussichtlich unerheblich. Aber das Peptid weist eine ausreichend große Struktur für die Analyse mit dem BIAcore X SPR-Instrument auf, dessen Nachweisgrenze etwa 200 Da ist. Deshalb wurde SPR verwendet, um die Absorptionseigenschaften von Fibronectin Adhesion-promoting Peptide auf der Polymeroberfläche zu messen. SPR-Sensorgramm der Adsorption von Fibronectin Adhesion-promoting Peptide auf einer begrenzten Poly(2-Vinylpyridin)-Oberfläche wurden bei verschiedenen Peptidkonzentrationen(in mg/mL) in HEPES-Puffer(pH 7) bei T = 25 °C durchgeführt: 0.1, 0.2, 0.3, 0.4, 0.5, 0.6, 0.8, 1.0 und 1.5. Die Flussrate war 40 μL/min.[2]

Außerdem fördert Fibronectin Adhesion-promoting Peptide konzentrationsabhängig direkt die Adhäsion, Ausbreitung und Migration von RCE-Zellen.[1]
